Acanthamoeba are abundant in a wide range of environments, and some species are responsible for cutaneous infections, keratitis, and granulomatous amoebic encephalitis (GAE). The conventional detection and isolation of amoeba from clinical and environmental samples involves sampling and culture on non-nutrient Ágar medium. Although efficient, this system requires several transfers in order to eliminate contaminants, and is not appropriate for the isolation of individual amoeba from samples with a biodiverse community. In this study we propose an alternative method for the isolation of monocystic clones of Acanthamoeba. The propose method involves sampling, enrichment, encystment induction, and direct cysts micromanipulation and culture on Ágar plates. |

Acanthamoeba ha sido aislada de numerosos hábitats incluyendo piletas de natación. Estudiar su distribución es importante ya que algunas especies causan enfermedad en el hombre. El objetivo del presente trabajo fue la búsqueda, aislamiento y caracterización de protozoos del género Acanthamoeba en piscinas cubiertas de la ciudad de Bahía Blanca, Argentina, en las cuatro estaciones del año durante 2007-2008. Se estudiaron 7 piscinas y en cada una se tomaron cuatro muestras: fondo, superficie, raspado de pared y para análisis bacteriológico. Las muestras se analizaron por observación directa y por cultivo en agar no nutritivo a 37 °C y a 42 °C. ABSTRACT Acanthamoeba is the most common free-living environmental amoeba, it may serve as an important vehicle for various microorganisms living in the same environment, such as viruses, being pathogenic to humans. This study aimed to detect and quantify human adenoviruses (HAdV) in Acanthamoebas isolated from water samples collected from swimming pools in the city of Porto Alegre, Southern Brazil.
